Siviilielämään vetäytynyt suupaltti Deadpool on masentunut automyyjä, joka potee keski-iän kriisiä. Kaikki muuttuu, kun Aikavarianssivirasto värvää Deadpoolin, jonka tehtävänä on pelastaa hänelle tärkeä aikajana. Apuunsa Deadpool saa vastahakoisen Wolverinen, ja vastustajakseen puolestaan mystisen Cassandra Novan.
